I am thinkng of taking a frame of bees from a very strong hive and adding it to another that needs building up. Uniting with a spare brood box and newspaper between the two. Do I need to stop the workers migrating upwards into the top box?
 
Not sure what you are planning here.
If you take a frame of emerging brood just shake the bees off and move it to the other hive as long as you are not risking disease transmission.
If uniting two colonies ensure one is queenless and unite. The point is to get the bees to mix but newspaper uniting as long as one half is queenless is straightforward. A lot on here just use air freshener and no paper.

As Colin says, I use air freshener. Quick squirt and add.you will get all sorts of problems with bees going into the spare brood box if you use newspaper. That is fine if you are uniting two large colonies but one frame? Air freshener route

Ah I get it know. Shake the fliers of a well laid frame and put it in place of an empty frame. No uniting tactic needed. Also ensures you don’t move a Queen by mistake.
