No to both questions. In the confocal microscope out of focus light is blocked with a pinhole. I set the z dimensions and how many steps or focus planes and how thick they should be, then it acquires all the slices. In Image J I deconvolved the stack to increase the resolution.

Just for your interest, this is a confocal stack of some pollen grains found in my honey. The stack has been average projected to form the 2D image after deconvolution in Image J.
The pollen was then artificially coloured using Photoshop.

Hi all,
I have had enough of the standard honey bucket gate valves (they leak all around the opening when using) and would like to put a lever style metal one in for next year.
Would any type of gate valve made with stainless steel grade 304 be acceptable? Anybody already done this?
Is it that you want to clean the slides to look at the specimens that are on them already? A wipe with some alcohol will remove oil etc.
If it is to recycle the slides (permanent mounts) please don't bother. I'd rather dry and reuse teabags.
